The primary benefit of the ActiveState distribution is that it is bundled with the Win32 software from Mark Hammond. This is described in Mark's book, "Python Programming On Win32: Help for Windows Programmers" (see http://www.amazon.com/Python-Programming-Win32-Windows-Programmers/dp/1565926218/ref=sr_1_1?ie=UTF8&qid=1304369987&sr=8-1). This is a good book, though it could certainly benefit from an update.

Windows Pythonistas might also want to look at IronPython, which integrates with .NET.
